Variasi Suhu Inkubasi Mempengaruhi Jumlah Sel Vegetatif dan Spora Bacillus Sphaericus

Aplikasi Bacillus sphaericus di lapang ditentukan oleh adanya inklusi kristal pada spora Bacillus sphaericus dan semua strain Bacillus sphaericus yang bersifat toksik terhadap nyamuk. Jumlahinklusi kristal pada spora dan sel vegetative Bacillus sphaericus sangat dipengaruhi oleh lingkungan inkubasinya, salah satunya adalah suhu inkubasi.Penelitian ini bertujuan untuk mengetahui pengaruh variasi waktu inkubasi terhadap jumlah sel vegetatif dan spora Bacillus sphaericus” pada media NYSM padat dan media NYSM cair.Penelitian ini merupakan penelitian Pre eksperimental di laboratorium, menggunakan 10 macam perlakuan variasi waktu inkubasi yaitu suhu 37 oC selama 1 hari, 2 hari, 3 hari,4 hari,5 hari, 6 hari, 7 hari,8 hari,9 hari dan 10 hari pada media NYSM padat dan NYSM cair. Data yang diambil yaitu jumlah sel vegetative Bacillus sphaericus dan jumlah spora Bacillus sphaericus Data dianalisisis secara deskriptif. Hasil penelitian menunjukkan hari pertama dan kedua (1 hari dan 2 hari) rerata jumlah sel vegetative 85,7 sel dan spora 14,3 spora. hari ke 3 sampai dengan ke -10 secara berurutan jumlah sel vegetative adalah 60,7; 36,4 ,18,4 ;10,4, 8,4;7,7dan 0. Jumlah Spora Bacillus sphaericus secara berurutan mulai hari ke-3 sampai dengan hari ke-10 adalah 39,4 ,63,7 ,81,7, 89,7, 91,7 ,92,3 dan 100. Kesimpulan yaitu semakin lamanya waktu inkubasi menyebabkan jumlah sel vegetative semakin berkurang dan jumlah spora semaki bertambah
